Project summary:
Self-immolative (SI) linkers are chemical constructs that undergo irreversible defragmentation upon external stimuli, releasing a cargo (a leaving group) which is crucial for various applications such as drug delivery and the construction of innovative materials. The main goal of the Ph.D. thesis is to search for novel phosphorus-based SI linkers bearing the following features: a) fluorescent tag suitable for biological response monitoring; b) one or two drugs as a payload; c) reactive moiety suitable for bio-conjugation or peptide stapling; d) alteration of features mentioned above. Linker development will consist of synthetic work – using organic synthesis methodology, application of photocage triggers, and 31P NMR irradiation experiments. Optimized SI linkers will be applied in various biological experiments or the delivery of drugs.

Expected/required skills:
A good feeling for the organic synthesis, knowledge of phosphorus chemistry is an advantage, good communication skills, and a positive mood in dealing with unexpected chemistry issues.
